You SHOULD be able to uncap that line. Since the pump is creating positive pressure on the front and negative on the back and water weight is keeping air from invading the system, you shouldn't have a geyser. With that said, is there room for air in that tube? If there isn't, take a syringe/turkey baster/straw (um ... ) and drain some room so there is an air gap at the top of the bleed/fill line. Keep the system running for a while, and eventually your bubbles will bleed out (bang on your system and tip it around to help them work free ... just don't cause your trapped air to get reconsumed into the loop). It is damn near impossible to fill a system without any bubbles in it.
