Will the orientation of this poly T work with this T-line? My radiator will be placed on the bottom of my stacker case and the barbs to the radiator will be to the front of the case. Due to my pump being close in proximity, I was thinking of setting it up as dipicted in the illustration. The short end of the poly t will be facing towards the pump inlet, and the other ends of the poly t will be right above the barb to the radiator. Sorry for the crude pic.

I wouldn't have the T in that direction because it will be restrictive due to 90 bend to pump and also the rad before the pump.

You will get better performance placing loop gpu>pump >rad> Tline> cpu >gpu.

you can mount the rad standing up or mount pump on the mobo tray if you want to place rad on the floor above the vent.
